菌株AVB101菌株 AVB101,一种大肠杆菌 B 菌株(hsdR、lon11、sulA1),含有 pBirAcm,一种 IPTG 诱导型质粒,其中含有工程化到 pACYC184 中的 BirA 基因。它与大多数克隆载体兼容,并用氯霉素 (10 µg/ml) 维持。该菌株被推荐用于蛋白质表达,因为它生长旺盛且不存在 OmpT 和 Lon 蛋白酶。Avidity, LLC 提供三种形式的菌株 – 甘油原液 (AVB101)、化学感受态细胞 (CVB101) 和电感受态细胞 (EVB101)。

BirA生物素蛋白连接酶(EC 6.3.4.15)激活生物素形成5'-腺苷酸生物素,并将生物素转移至生物素受体蛋白(例如AviTag™肽)。它还起着生物素操纵子阻遏物的作用。该蛋白质由birA基因编码。

BirA生物素蛋白连接酶(EC 6.3.4.15)通过ATP中间体(生物素5'-腺苷酸)以高效,针对性的方式将d-生物素共价添加到生物素受体肽/蛋白质上。酶促生物素化蛋白的下游应用是多种,重要和强大的。通常利用*的生物素-亲和素/链霉亲和素相互作用来进行亲和层析或将蛋白质固定在表面或底物上。通过抗生物素抗体或抗生物素蛋白/链霉亲和素-报告酶偶联物(-HRP,-碱性磷酸酶)或荧光探针进行蛋白质检测成为可能。生物素化的MHC分子的多聚体形式是免疫生物学中流行的工具。

生物素标记后可以采用SABC(Streptavidin-Biotin Complex)法进行检测;也可以使用辣根过氧化物酶标记的Streptavidin或碱性磷酸酯酶标记的Streptavidin来识别生物素,后续用辣根过氧化物酶或碱性磷酸酯酶对应的检测体系进行显色反应或发光反应等。
本抗体为用纯化的兔IgG(H+L)免疫山羊,然后用亲和纯化柱对获得的抗血清进行纯化,并经过小鼠IgG等吸附纯化和生物标记的优质二抗。对小鼠等的血清蛋白几乎没有结合能力(Minimal Cross-reaction to Mouse)。
本生物素高效标记山羊抗兔IgG(H+L)每摩尔IgG结合多摩尔的生物素,能结合多个streptavidin或avidin分子,在SABC或ABC等检测系统中,能有效放大信号,提高检测灵敏度。
本生物素高效标记山羊抗兔IgG(H+L)用于Western、免疫细胞化学、免疫荧光、免疫组化和ELISA的推荐稀释比例参考上表,实际实验操作过程中需根据抗原和抗体的具体情况适当调节二抗的稀释比例。
本抗体如果用于常规的Western检测,以每次检测需10毫升1:2000稀释的二抗计算,可以检测100次;如果用于常规的免疫细胞化学或免疫组化检测,按照每样需要100微升1:100稀释的本抗体计算,可以检测500个样品。
