凝血酶原
的蛋白水解激活丝氨酸蛋白酶α-凝血酶是通过酶原,凝血酶原的蛋白水解激活而产生的。酶复合物凝血酶原酶催化凝血酶原中两个肽键的蛋白水解,从而产生一个NH2末端衍生的F1.2区和异二聚体α-凝血酶。α-凝血酶由“ A”链(Mr = 6000)组成,该链通过单个二硫键与“ B”链(Mr = 31,000)共价连接。

α-凝血酶是通过酶原凝血酶原的蛋白水解激活而产生的高度特异性的丝氨酸蛋白酶(1)。在凝结过程中,凝血酶裂解纤维蛋白原形成纤维蛋白,导致凝结的终步骤,即形成纤维蛋白凝块。凝血酶还负责前因子V和VIII因子的反馈激活。还已经报道凝血酶激活因子XIII和血小板,并且还起血管收缩蛋白的作用。凝血酶的促凝血活性通过两种方式被阻止:1)被肝素辅因子II或抗凝血酶III /肝素复合物抑制。或2)与血栓调节蛋白形成复合物。凝血酶/血栓调节蛋白复合物的形成导致凝血酶无法裂解纤维蛋白原并激活因子V和VIII,
凝血酶是由NH 2末端“ A”链(Mr = 6,000)和COOH末端“ B”链(Mr = 31,000)组成的两条链酶,它们通过单个二硫键共价结合。人凝血酶比牛凝血酶短13个氨基酸,这是由于人蛋白上的凝血酶裂解位点不存在于牛蛋白中。
凝血酶还用于细菌中表达的融合蛋白的位点特异性切割(9-11)。凝血酶敏感位点被掺入目的重组蛋白和促进纯化和/或表达的肽或蛋白之间。通过用凝血酶裂解从表达的中释放靶蛋白。然后可以通过亲和层析容易地除去凝血酶。
人,牛和小鼠的凝血酶是按照Nesheim等人的描述,使用Lundblad程序(1)的改进方法,从纯化的凝血酶原中制备的。(2)。凝血酶以50%(体积/体积)甘油/水的形式提供,应储存在-20oC下。通过SDS-PAGE分析确定纯度,并在凝血酶特异性凝血测定中测量活性,并与标准NIH凝血酶进行比较。凝血酶也可通过DFP,FPRck或生物素化FPRck阻断活性位点。
融合蛋白的裂解
除了其在凝血研究中的广泛应用,凝血酶还可用于融合蛋白的位点特异性裂解。凝血酶敏感位点被掺入目的重组蛋白和促进纯化和/或表达的肽或蛋白之间。通过用凝血酶裂解从表达的释放靶蛋白。然后可以通过亲和层析容易地除去凝血酶。批次间的一致性确保每次都能获得可重复的结果。对于涉及细胞培养的实验,请与我们联系以讨论用于细胞培养的定制低内毒素批次。

【生理作用】当循环凝血因子在暴露的血管外组织与组织因子相接触时,凝血酶会在组织上聚集。凝血酶通过激活血小板,催化纤维蛋白原转化为纤维蛋白,促进血稳定而在血栓性疾病的引发和传播上有着核心作用
α-凝血酶(α-Thrombin)是一种高度特异性的丝氨酸蛋白酶,由凝血酶原(prothrombin,II因子)经蛋白水解活化而来。它在凝血过程中起着非常重要的作用,不仅能够促使纤维蛋白凝块的产生,还负责提供反馈信号来激活前辅因子:V因子和VIII因子。也可以激活XIII因子和血小板,或者用作血管收缩剂。
在体内,活化的X因子(Factor Xa)切割凝血酶原,释放出活性肽并将凝血酶切割成具有催化活性的α-凝血酶。α-凝血酶由一条轻链(A chain)(Mw ~6,000)和一条重链(B chain)(Mw ~31,000)通过二硫键连接而成。某些情况下,α-凝血酶会发生自溶生成β-凝血酶和γ-凝血酶。β-凝血酶由对α-凝血酶A链水解并切割含有B链糖基化位点的小片段(B1,B2)组合而成。
除了用于凝血研究之外,α-凝血酶还常用来位点特异性切割融合蛋白。基因重组时将凝血酶识别位点插入在目的蛋白与利于后续纯化和/或表达的多肽或者蛋白之间,通过凝血酶切割表达的重组子即可释放目的蛋白。凝血酶本身可通过亲和层析技术快速去除。
作为生物活性物质的凝血酶:是一种在人体抗凝,促凝的凝血级联反应中有着重要作用的生物活性蛋白。

Prepared from Bovine Factor IX by activation with Bovine Factor Xla, this Bovine Factor Xla is removed after activation. Complete activation is observed by SDS-PAGE. The Factor Xla activates FIX in a two-step reaction. In the first step, an internal Arg-Ala bond is cleaved, and in the second step, an Arg-Val bond is cleaved. The second cleavage leads to the liberation of an activation peptide from the NH2-terminal portion of the heavy chain to produce factor IXaβ.
