NFAT响应性荧光素酶报告基因Jurkat T稳定细胞系来自人T淋巴细胞,并在NFAT响应元件的控制下稳定表达萤火虫荧光素酶报告基因。该细胞系是用于监测由刺激治疗,强制基因表达和基因敲除引发的钙信号通路的激活的理想细胞模型。
NFAT是转录因子家族,在免疫反应以及心脏,骨骼肌和神经系统的发育中起重要作用。NFAT受钙信号调节。通过磷酸酶钙调神经磷酸酶的钙激活,NFATc蛋白从细胞质转移到细胞核中,与其他蛋白质配合以介导基因表达。NFAT的核进口被PKA和GSK3等激酶阻断。NFATs也涉及乳腺癌。Signosis已经建立了已经用NFAT-荧光素酶报告基因构建体稳定转染的NFAT荧光素酶报道细胞系。通过荧光素酶的分析,细胞系可用于监测由刺激,复合治疗,强制基因表达或基因敲低引发的NFAT活性的细胞变化。
通过使用含有NFAT结合位点,萤火虫荧光素酶编码区上游的zui小启动子以及潮霉素表达载体,然后潮霉素选择的pTA-NFAT-荧光素酶报道载体转染建立细胞系。随后对潮霉素抗性克隆进行PMA +离子霉素诱导的荧光素酶活性筛选。
TF荧光素酶报告原理。 TF荧光素酶报道稳定细胞株利用人工启动子构建体来驱动荧光素酶表达。启动子区域可以由顺式元件TF结合位点的多个重复序列,来自已知TF下游基因的启动子区域的DNA片段或含有推定/已知的TF结合位点的DNA片段组成。TF可以通过细胞外刺激或通过细胞内信号传导途径激活几种方式。一旦激活,TF易位于细胞核,并经常与相关因子相互作用以驱动基因表达。一旦荧光素酶表达,其可以在酶测定中产生光,并且测量的光量与TF活化的水平正相关。
