产品描述：

EZ Mark彩色预染蛋白分子量标准包含了从15k到130k共8种纯化的预染蛋白质(15,20,25,35,50,70,100,130kD)，其中70kD条带为橙色，其余条带为蓝色，适合作为SDS-PAGE或Western的蛋白质分子量标准。

本彩色预染蛋白质分子量标准已经配制在1×SDS-PAGE上样缓冲液中，可以直接使用，无需煮沸。

根据上样孔的大小，本彩色预染蛋白质分子量标准通常每次上样5-10微升（5×1.5mm胶孔5ul足够），即可在电泳时、电泳后或转膜后观察到非常清楚的蛋白条带。

使用方法：

1. 室温下解冻后轻轻混匀或者是用移液枪缓慢吹打均匀，不要煮沸。

2. 取本产品5ul与实验样品同时进行聚丙烯酰胺凝胶电泳；建议有条件的实验室在初次使用本产品时可以根据自身的实验条件和实验习惯通过预实验确定合适的上样量，这样可以节约成本，同时获得效果更佳的实验图片。

3.未使用的蛋白分子质量标准保存于储存条件或短期在4℃放置。

运输与保存方法：

冰袋运输。-10℃到-30℃条件下储存。

注意事项：

  1. 通常电泳至蓝色的溴酚蓝基本上到达凝胶底部或预染分子量标准充分展开时停止电泳。
  2. 购置冻干Marker产品的客户，如果需要自己配制冻存液，可参考配方：【储存液】67 mM Tris-HCl, pH7.5, 20 mM DTT, 5mM EDTA, 2 % (W/V) SDS, 33 % (W/V) Glycerol, 0.02 % (W/V) proclin300.
  3. Marker上样多或者某些特定情况下，35和50k之间会显示的弱带为40k。

产品描述：

细胞转染液（核心成分为线性聚乙烯亚胺，也称作LPEI），是新一代的转染试剂，带正电的聚合物与核酸带负电的磷酸基团 形成带正电的复合物后与细胞表面带负电的蛋白多糖相互作用，并通过胞吞作用进入细胞。除了具有阳离子脂质体（如：Lipo2000,3000）的转染效率高，操作简单，适用范围广，重复性好等特点外，还具有在体内，转染效率高，细胞毒性低等特点。

EZ Trans是一种具有较高的阳离子电荷密度的有机大分子，每相隔二个碳个原子都是质子化的氨基氮原子，使得聚合物网络在任何pH下都能充当有效的“质子海绵”体。其转染性能好于树枝状聚合物，而且它的细胞毒性低。

使用方法：

瞬时转染方法

1. 接种细胞：转染前一天，用胶原酶消化细胞并计数（不建议用胰酶）。调整细胞浓度，将细胞铺入细胞培养的器皿，每个孔置入的细胞量应能使转染时细胞汇合度达到 70~80%。

2. 准备 DNA-PEI 复合物： DNA、EZ Trans 试剂和稀释剂在进行以下步骤前需先使其升至室温。依据下表所示，用 Opti-MEM ITM（Invitrogen）或其他适合的无蛋白培养基稀释适量 DNA。用同样的培养基稀释EZ Trans 试剂。每 1 μg DNA 需用2-5 μL EZ Trans转染试剂。一边轻轻涡旋装有 DNA 溶液的试管，一边将稀释的EZ Trans转染试剂滴加至试管中（注意：请勿颠倒添加顺序）。充分混匀后，室温静置 10~25 min 以形成DNA-PEI复合物。当溶液体积较大时，请用圆底聚丙烯管，例如 Falcon 5 mL /14 mL 离心管。

3. 转染细胞：直接向每个孔中加入 DNA-PEI复合物并轻轻涡旋培养板/培养皿。在无血清条件下转染时，去除生长培养基，替换成无血清培养基，然后滴DNA-PEI复合物。转染 3 h 后，添加1/2体积的包含 30%血清的生长培养基。

4. 孵育细胞和分析结果：在 CO2培养箱中 37℃下孵育细胞至可以分析检测。转染后zui快 7h 即可检测到转入基因的表达。请自行确定检测时间。?

稳定转染方法

1. 接种细胞：转染前一天，用胰酶消化细胞并计数。调整细胞浓度，将细胞铺入细胞培养的器皿，总体积如表 1 所示，每个孔置入的细胞量应能使转染时细胞汇合度达到 70~80%。

4. 孵育细胞和分析结果：在 CO2培养箱中 37℃下孵育细胞至可以分析检测。转染后zui快 7h 即可检测到转入基因的表达。

5.转染 24 h 后，将细胞传代至新鲜的生长培养基中（将细胞稀释 10 倍以上），在 CO2培养箱中 37℃孵育隔天。第二天加入与转染抗性基因相匹配的筛选药物。约 1~2 周可筛选到耐药性克隆，在这期间需经常更换含筛选药物的生长培养基。

运输与保存方法：

常温运输，4℃保存，保质期12个月。

使用注意事项:

质粒质量：请务必使用高质量转染级无内毒素质粒。通过 260 nm 光吸收测定 DNA 浓度，260 nm / 280 nm 比值确定 DNA 纯度（比值应该在 1.8~2.0 的范围之内）。如有可能，请通过琼脂糖凝胶电泳检测质粒的完整性。

细胞条件：使用适当保存和经常传代的健康细胞。确保培养基没有被细菌、真菌或支原体污染。如果细胞是近期复苏的液氮冻存细胞，请在转染前至少传代两次。建议使用GIBCO或者李记生物的胎牛血清培养细胞。?

特别提醒:

1. 对于某些类型的细胞如 HEK-293、HEK293T、NIH/3T3 和 COS 细胞，在转染前两天铺板可显著提高转入基因的表达水平。如果选择转染前两天铺板，可适当降低铺板密度，以确保转染时细胞的汇合度仍为 70~80%。

2. 对于接触抑制敏感的细胞，可适当降低铺板密度。

3. 即使在有蛋白（如 10%的血清）存在的情况下，DNA-PEI复合物仍能转染细胞，但是 DNA-PEI复合物必须在无蛋白存在的条件下形成。

4. 对大多数细胞来而言，每 1 μg DNA 使用 3.0 μLEndoFectinTM 试剂都能获得较高转染效率。使用者也可尝试每 1 μg DNA 使用 1~4 μL 体积线性PEI转染试剂进行优化。

使用方法

1. 取2支小烧杯（小试管亦可），在*个小烧杯中加入试剂盒中的EZ Protein anyKD PAGE 浓缩胶A液1.0ml + EZ Protein anyKD PAGE 浓缩胶B液1.0ml；在另一个烧杯中加入EZ Protein anyKD PAGE 分离胶A液2.5ml + EZ Protein anyKD PAGE 分离胶B液2.5ml；

2. 在5ml 的分离胶溶液中加入10%APS 50ul，轻轻搅拌使其混匀，避免产生气泡；

3. 在凝胶模具中灌入适量混匀的A液（对于 mini-gel，凝胶液加至约距前玻璃板顶端1.5cm或距梳齿约0.5cm即可）；

4. 在2ml 的浓缩胶液中加入10%APS 20ul，轻轻搅拌使其混匀后直接灌入凝胶模具中A液的上层（不需要等待A液凝固）；

5. 将梳子插入凝胶内，静置10~15分钟，等待浓缩胶聚合；

6. 待凝胶聚合后，小心地拔出梳子，以免破坏加样孔；

7. 调整电泳仪为恒压 300v，进行电泳操作，约30min完成蛋白电泳实验。（300v为绝大多数电泳仪设计可承受电压，正常使用不会损坏仪器。如有担心可以适当降低电压至250v，对电泳速度不会有明显影响。）

注意事项

1. 本试剂盒能够一次性完成浓缩胶和分离胶的制备不会发生浓缩胶和分离胶相互混合的情况；

2. 由于本产品具有快速凝胶的特点，在凝胶配制过程中遵循操作说明使用，不要将分离胶灌入凝胶模具后，才开始配制浓缩胶（间隔时间过长可能会导致分离胶部分聚合），这样灌制浓缩胶时容易引起分离胶与浓缩胶分界面不平，影响电泳效果；

3. 在灌胶过程中使用小烧杯或者小试管作为配胶容器，按照操作说明配制好分离胶和浓缩胶后立即先后将分离胶和浓缩胶灌入凝胶模具中。不建议使用使用移液器缓慢加入可能会导致分离胶和浓缩胶分界面不平。

4. 需要一次性制备多块蛋凝胶时，为防止快速凝胶可以适当减少过硫酸铵的用量；

5. 本产品有针对性的耐高压凝胶系统，可以在常规电泳缓冲液中实现快速电泳的目的；

6. 如长期大量使用凝胶，可以用本试剂盒一次性配制多块凝胶，作为预制胶，放入加有少量电泳缓冲液的自封袋中，于2-8℃环境下长期保存使用；

7. 操作步骤中的凝胶制备是以Mini-gel的规格制定，如需要制备较大的凝胶可以适当调整试剂的用量；

8. 尽管本产品对各种原料已做了调整和修饰，极大的提高了实验操作的安全性，但是依然含有丙烯酰胺，因此操作时请穿着实验服并佩戴一次性手套。

INCELL EZ-CPZ™ 冻存液

​EZ-CPZ™是一种无血清、无蛋白的冻存液，不含任何不明确的或动物来源的成分，适宜于临床或特殊使用目的的细胞冻存（如无血清培养细胞）。冻存于该冻存液的人或其他动物细胞复苏后保持良好的活性、生长性及分化潜能。可以与细胞悬液以1：1混匀冻存，细胞无需离心。使用前添加DMSO至终浓度5%。

用途和方法

这包括来自多个人体实体组织，血细胞和肿瘤细胞的新鲜分离的细胞（表1）。可从类似的动物组织获得可比较的结果（表2）。从这些来源培养的细胞很少或没有传代培养，然后储存在EZ-CPZ TM中，在台盼蓝染料排除测定中具有从75％至90％或更低的低回收生存力。将来自贴壁或悬浮培养物的细胞与EZ-CPZ TM以1至10×10 6个细胞/ mL（取决于细胞类型）1或1轻轻混合，或其他优选的细胞密度不超过冷冻保存培养基总体积的20％。血清或血浆的添加是任选的，并且如果目标是维持确定的细胞系统则是有害的。在具有或不具有其自体（或异源）血浆或具有抗凝血剂的血清的培养基中新鲜收集的肿瘤细胞或血细胞也可以与EZ-CPZ TM以1：1混合。或者，EZ-CPZ TM可直接用作标准10％（v / v）DMSO冷冻保存介质，以重悬细胞浓缩物或颗粒。将冷冻管或袋子放入控制速率的冷冻容器或“先生”中。在-80℃下用异丙醇冷冻至少12小时，以每分钟约1度的速度冷却。然后将它们转移到-135℃冰箱或液氮冷冻机中。通过将冷冻管置于37℃水浴中约1分钟直至大部分培养基解冻，快速回收冷冻细胞。将细胞立即置于预热的细胞培养基中。此时细胞非常脆弱，必须轻柔处理。大多数细胞在培养基中生长几小时或过夜后从冷冻保存中恢复。虽然通常不需要，但是将一些细胞离心以除去冷冻保护剂培养基，然后轻轻地重悬于生长培养基中。

细胞系
INCELL专有的E10（抗HHV-8）杂交瘤，NCM460D™，NCM356D™，IBHK-4和其他标准细胞系已在EZ-CPZ™中成功并重复进行冷冻保存。它还被用于从许多物种和细胞类型中冷冻许多标准细胞系，并在其推荐的培养基中生长。一系列实例包括CHO，BHK，COS，293,3T3，U937，HT29和HL60。

M3base 基础培养液中，A型为含复合抗生素，适宜于有污染可能的细胞培养，但是对细胞有一定的伤害，F型不含抗生素；C型不含有酚红指示剂，适用于细胞标记实验，且酚红对部分细胞有生长影响及毒害作用。

M3：10为M3基础培养液添加10%胎牛血清，适宜于胚胎或成体干细胞培养、培养条件严格的间质细胞培养，贴壁细胞培养、多数肿瘤细胞的起始培养液，以及用于促进细胞株生长、分化的培养液等。

M3:D为培养液成分明确，不含任何不明成分生长因子及血清成分，可作为添加特定生长成分实验的对照实验培养液。

SMX为成分明确的生长因子复合物。

2、ACE™ 无血清培养液

ACE™是成分明确的，低蛋白 (<20mg/L)的无血清培养液,适宜于适应能力低的细胞培养，以获得良好的生存和生长能力。培养中可不需要二氧化碳, 减少了疫苗生产下游处理程序。同时不添加酚红，在杂交瘤细胞培养时比同类培养液产生更多的抗体。

 

三、冻存液及细胞处理液

1、CPZ™ 冻存培养液

​CPZ™冻存液含有临床级别的羟乙基淀粉、右旋糖苷及配方（生长因子，%10胎牛血清，冷冻保存剂）。适宜于包括神经细胞在内的多种人及哺乳动物细胞冷冻保存。使用前添加DMSO至终浓度5% to 10% (v/v),（根据冻存细胞类型确定浓度）。冻存细胞（含少量培养液）体积应少于冻存液体积的20%，细胞数通常为1 -25 x 106 cells/mL。将细胞轻柔重悬于冻存液后置于-80度冰箱至少12小时（放于程序冻存盒内使其温度以1度/分钟缓慢下降），然后转入–135度冰箱或液氮罐长期保存。

四、细胞株

1、NCM356细胞株（Normal-Derived Colon Mucosa Cell Line，正常的结肠粘膜细胞株）：

   来源于一名65岁男性黑人直肠腺癌患者的结肠粘膜上皮，未注入任何外源基因，表达细胞角蛋白、绒毛蛋白结肠上皮细胞抗原，不表达其它特定细胞株表面抗原。部分细胞表达黏液素合成酶及肿瘤相关抗原。不能在软琼脂上生长，无致瘤性。要求严格的生长条件，须在INCELL's 的M3:10TM 培养液上做长期培养，倍增时间32-35小时。可广泛用于肠道细胞的研究，如消化道感染疾病、细胞信号转导，细胞因子表达，维生素转运及基因调控。

3、IBHK-4细胞株（A Baby Hamster Kidney Derived Cell Line，幼仓鼠肾脏细胞）

   IBHK-4细胞株来源于仓鼠肾脏，常规于INCELL的ACE™培养基上悬浮生长，培养液需要无血清、无抗生素及动物源成分，培养时需要轻微摇晃。能在软琼脂上快速形成集落，因此有致瘤性。该细胞株可广泛应用于病毒培养，用于灭毒疫苗生产。
