异质核RNA（hnRNA）的微粒复合物是高Mr的RNA分子的异质混合物，其在蛋白质合成过程中在细胞核中发生，具有细胞特异性和异质性的蛋白质。蛋白质组分可能在加工过程中起作用。 hnRNA对mRNA的影响

免疫荧光方案 – 甲醛固定

   1.从Tcunit收集细胞，并用吸力从培养皿中取出培养基。
   2.用1x PBS洗涤并取出。
   3.在室温下在轨道振荡器上将细胞在预热（37℃）对甲醛中孵育12分钟。
   4.去除PFA并在室温下在1x PBS中的0.5％Triton X-100中孵育5分钟。
   5.准备封闭试剂，这也是抗体稀释剂。
   6.在室温下用1x PBS洗涤细胞2次，在轨道振荡器上洗涤4分钟/洗涤。
   7.在室温下用1％NCS和1x PBS封闭30分钟。
   8.准备一抗（50μl/盖玻片）和湿润染色室。
   9.在室温下用1x PBS洗涤细胞2次并短暂风干。
10.在室温下在黑暗的染色室中与一抗孵育1小时。在此期间准备二抗。
11.用1x PBS洗涤细胞5次（每个烧杯更换5个烧杯/ 5个计数）
12。在室温下在黑暗的染色室中与第二抗体孵育1小时。
13.用1x PBS洗涤细胞5次。
14.登上达皮。

溶液（染色当天新鲜制备）。

    * 1x磷酸盐缓冲盐水。
    *封闭试剂：1x PBS中1％NCS（使用新鲜的10x PBS）。
    *固定液：3.5％对甲醛。

1.75g PFA在20ml d.H2O中加5滴1M NaOH。在50-60℃的热板上搅拌直至溶解。加入4滴IN HCl并检查pH指示条。PH值应为7.4。完成体积，d.H20至25ml，加入25ml 2xPBS。在添加到盖玻片之前检查pH值。

免疫荧光方案 – 甲醇/丙酮固定

   1.从TCunit收集细胞，并用吸力从培养皿中取出培养基。
   2.用1x PBS洗涤并取出。
   3.用冷甲醇：丙酮1：1在冰上固定细胞10分钟。
   4.制备阻断试剂，这也是抗体的稀释剂。
   5.取出固定剂并在室温下用1x PBS洗涤细胞3次，在轨道振荡器上洗涤4分钟。
   6.在室温下用1％NCS和1x PBS封闭30分钟。
   7.准备一抗（50？l /盖玻片）和湿染色室。
   8.在室温下用1x PBS洗涤细胞2次并风干约7分钟。
   9.在室温下在室温下与一抗孵育1小时，在染色室中避光。在此期间准备二抗。
10.用1x PBS洗涤细胞5次（每个烧杯更换5次烧杯/ 5次计数）
11。在室温下，在染色室的黑暗中与第二抗体孵育1小时。
12.用1x PBS洗涤细胞5次。
13.登上达皮。

在TOPGAL小鼠中，口服IQ1 (72 or 144 mg/kg)抑制β-catenin/p300相互作用，破坏了小鼠肺上皮在子宫内和离体的远端分支，使肺上皮近端化。^[²^]
