标签抗原由于其分子量小,对融合蛋白的活性没有影响等特点,已被广泛应用于重组蛋白在后续免疫印迹、免疫沉淀和免疫技术的检测中。Flag标签是重组蛋白研究中应用较为广泛的一种,该标签系统利用一个短的亲水性八氨基酸肽(DYKDDDDK)融合到目标蛋白的N-端或者C-端,且该标签可被后续肠激酶去除。Flag抗体可以用于检测和Flag标签融合表达蛋白的表达、细胞内定位,以及纯化、定性或定量检测Flag融合表达蛋白等。
