dsDNA BR Assay Kit 是一种简便、灵敏、准确、宽范围的双链 DNA（dsDNA BR)荧光定量检测试剂盒，在2~1000 ng 区间具有良好的线性关系。本试剂盒包含荧光检测试剂、缓冲液及相关的 dsDNA BR 标准品，使用前先将荧光检测试剂用缓冲液稀释成工作液，然后加入待测 dsDNA 样品，制作标准曲线后，使用荧光酶标仪或 Qubit®荧光仪进行读数。该产品对常规的污染物如蛋白质、盐类等具有较好的耐受性。

注意事项

1）荧光染料均存在淬灭问题，请尽量注意避光，以减缓荧光淬灭；

2）检测试剂 dsDNA BR Buffer 略起泡，使用时上下颠倒混匀，避免剧烈震荡；

3）dsDNA BR 标准品，每次使用轻柔震荡或颠倒混匀，瞬时离心数秒收集管盖及管壁上的液体；

4）为保证定量结果的精确，请使用校准后的移液器操作，样品浓度较低时请增大待测样品投入体积；

5）检测工作液请现用现配，当天使用完毕，使用前请用标准品校准。

6）为了您的安全和健康，请穿实验服并戴一次性手套操作；

7）本产品仅用作科研用途！

 

实验步骤

使用 Qubit荧光仪进行 dsDNA BR 定量检测分析

dsDNA BR Assay Kit

1.实验准备

1）在使用前，将试剂盒中的各组分恢复至室温（室温放置约半小时）。检查 dsDNA BR Reagent（组分 A）是否有沉淀，若有沉淀物，可将该试剂至于 37℃水浴锅中温育，并轻柔混匀直至沉淀物完全溶解。

准备足够量 0.5 mL PCR 薄壁管并标注。请勿在 PCR 管侧壁标注，以免影响荧光信号采集。

2.配制检测工作液

在避光塑料容器中，使用 dsDNA BR Buffer 按比例将适量 dsDNA BR Reagent 稀释至 1×（例：取 1 μL dsDNA BR Reagent， 加入 199 μL dsDNA BR Buffer），上下颠倒混匀。工作液现用现配，每次配制检测工作液时要使用洁净的容器。

3.配制待检样品

1）配制待检标准品 1 和标准品 2。取 190 μL 检测工作液至标准品 PCR 管中，分别加入 10 μL dsDNA BR Standard 1 和 dsDNA BR Standard 2 至相应标记的标准品 PCR 管中，轻柔涡旋振荡 2-3 sec，尽量避免气泡产生，瞬时离心数秒。

2）配制待检样品。取 180-199 μL 检测工作液至样本 PCR 管中，分别加入 1-20 μL 待检样本，使 PCR 管中每个样本终体积为 200 μL，轻轻涡旋振荡 2-3 sec，尽量避免气泡产生。

4.检测

1）将所有待检 PCR 管置于室温避光孵育 2 min。

2）按照 Qubit荧光仪的操作说明，选择 dsDNA BR 检测程序，使用待检标准品校正荧光曲线后进行待检样品的浓度测定。

预期用途
该试剂盒是一种夹心酶免疫分析法，用于体外定量测定人体AIF
血清、血浆、组织匀浆和其他生物液体。仅供研究使用。不适用于
诊断程序。

需要但未提供的材料
1.带450±10 nm滤光片的微孔板阅读器。
2.单通道或多通道移液管，具有高精度和一次性j端。
3.微量离心管。
4.去离子水或蒸馏水。
5.用于吸干微孔板的吸水纸。
6.洗涤液容器。
7.0.01 mol/L（或1×）磷酸盐缓冲盐水（PBS），pH值7.0-7.2。
存储
所有试剂应按照瓶上的标签保存。校准器、检测试剂A、，
检测试剂B和96孔条形板在接收时应储存在-20°C下，而其他试剂则应储存在-20°C下
温度应为4°C。未使用的板条应保存在密封袋中，并向其提供干燥剂
尽量减少暴露在潮湿空气中。打开的测试套件将在1个月内保持稳定，前提是将其储存为

校准器

Eagle Biosciences S100B ELISA测定试剂盒是用于定量测定人血清或血浆中S100B浓度的免疫酶学比色法。S100B ELISA测定试剂盒仅供研究使用。

测定原理

S100B ELISA测定试剂盒基于S100B通过两种抗体的结合，一种固定在微孔板上，另一种与辣根过氧化物酶（HRP）结合。该测定是两步结合方法，并且在每个孵育步骤之后，通过简单的固相洗涤进行结合/自由分离。然后，结合级分中的酶HRP与底物（H 2 O 2）和TMB底物反应，并且当添加停止溶液（H 2 SO 4）时，发展出变成黄色的蓝色。颜色强度与样品中的S100B浓度成比例。通过校准曲线计算样品中的S100B浓度。

测定背景

S100是属于EF-手钙结合蛋白的S100 /钙调素/肌钙蛋白C超家族的20kDa蛋白质。S100zui初是从人脑分离出来的，并被认为是胶质细胞特异性蛋白质（1）。今天，已经根据结构和功能相似性确定了S100家族的20种单体（2,3）。大多数S100蛋白以二聚体形式存在并以细胞特异性方式表达。被称为S100α和S100β（4）的S100单体中的两种在物种之间是高度保守的，并且在中枢神经系统胶质细胞和某些外周细胞中被发现为同型（ββ）和异二聚体（αβ）。雪旺氏细胞，黑素细胞，脂肪细胞和软骨细胞（5）。S100αβ和S100ββ也存在于恶性组织中，zui显着的是黑素瘤，而在胶质瘤，甲状腺细胞癌和肾细胞癌中也较少。血清中S100B（如S100αβ和S100ββ单位）的测定已被证明在诊断为恶性黑素瘤的患者的预后和治疗监测中具有临床意义（6-9）。研究还表明，S100B可能有助于治疗患有脑损伤的患者。创伤性头部损伤，围产期窒息，心脏骤停，心脏手术和中风（10-13）。
